摘  要:为研究人表皮角质细胞(EKC)的迁移活动,应用显微摄像方法在金颗粒吞噬玻片上观察,发现EKC的迁移运动可区分为缓慢平移、多向快速运动及飘移3种形式。非活化EKC在非调理情况下仅有极微弱的缓慢平移,而基质蛋白的调理作用可使其出现持续时间较短、相对活跃的迁移活动;活化的EKC在非调理情况下也有比较活跃的迁移活动,基质蛋白调理可使其迁移活动的3种形式表现得很活跃,运动持续时间最长。结论认为,活体创面具备活化及基质蛋白调理的条件,体外观察到EKC的3种迁移活动形式,可出现在活体创面重上皮化的修复过程中。We have developed an in vitro assay to observe the migrational movements of human epidermal keratinocytes (HEKCs) using microvideography. The migrational movements of HEKCs exhibited on gold particle-coated coverslips can be differentiated into a gentle translational migration, a random migration and a drift migration. In our experiments, we have found cultured,activated HEKCs move a little more actively than non-activated HEKCs freshly harvested from skin, and the opsonification of extracellular matrix (ECM:FN and type Ⅰ Collagen) can promote the migrational movements of the activated or non- activated HEKCs. Epidermal cells from the edges of wounds (full or non full thickness cutaneous wounds) and epithelial cell remnants from beds of wounds (non full thickness cutaneous wounds)are also acted by the two important factors: activation (wounding) and opsonification of ECM which is present in a large quantity in wound areas, but we still couldn't make the conclusion that the three migrational movements mentioned above are the real migrational movement model of HEKCs in vivo during the process of reepithelization, so there isroom for further experimental demonstration.
