摘  要:探索海绵理植的方法,获取烧伤创面的组织渗液,以建立一个适合烫伤创面愈合研究的动物模型。将经过预处理的PUA海绵理植大鼠背部二侧皮下,再予背部烫伤,制成海绵理植的大鼠烫伤模型(SB组);同时设立单纯海绵植入而不予烫伤的大鼠对照组(S组)。收集两组大鼠创面海绵渗液,观察其渗液量、渗液细胞量、渗液细胞TGFα、TGFβ的mRNA表达,以及渗液中IL-1、IL-6及TNF的水平。结果表明:烧伤3d后,SB组大鼠创面渗液的细胞量、IL-1、IL-6、TNF及生长因子水平大都明显高于对照组。这一差异无疑是由烧伤因素造成,因而能反映烫伤创面愈合的规律,是个较理想的研究模型。SD rats undergone dorsal skin incision with subcutaneous implantation of sterile, moist polyviny1 sponges under aseptic condition were divided into two groups. The control group of rats (group S) had been implanted sponges only, while the experimental group (group SB) a thermal injury that was inflicted immediatey after sponge implantation. Both groups of animals were killed postburn, and the sponges were removed and squeezed to collect the fluid of the wound. The cell - free fluid was used for cytokine(IL- 1, IL- 6 TNF) assay and the deposited cells used for checking mRNA expression of peptide growth factors (TGFα, TGFβ, PDGF). Our data show the following. The number of infilammatory cells, mRNA expression of growth factors and cytokine level of group SB are higher significantly than those of group S postburn. It is clear that these alterations between two groups are mainly attributed to burn trauma itself So we think this model provides a suitable basis for studying kinetics of various regulators in response to burn wound healing.
