摘  要:应用~3H-胸腺嘧啶核苷参入法,测定可溶性生长因子(EGF、PDGF、TGF-β)等对体外培养人表皮角质细胞(EKC)增殖的影响。结果发现EGF在1~5ng/ml时,EKC增殖迅速;浓度再升高时,EKC的CPM值增加趋向平缓。胰岛素可促使EKC增殖;当与EGF合用时,增殖呈明显的协同作用。EGF与PDGF和(或)TGF-β配伍时,代表细胞增殖的CPM值增量不明显,甚至下降。PHA在低浓度时可促进EKC增殖,高浓度时抑制,呈典型的剂量-效应曲线;PHA与EGF配伍,EKC增殖反应表现出一些复杂而有规律的变化。结果提示:1.可溶性生长因子确能促进体外培养EKC的增殖。2.EGF对EKC的增殖作用可受其他生长因子、胰岛素等外来因素的介入而发生改变。The writers have investigated the effects of soluble growth factors (EGF, TCG-βand PDGF), insulin and PHA on the proliferation of EKCs in vitro using a ~3H-thymidine incorporation assay. There was a period of rapid EKC proliferation stimulated by EGF at the concentration of 0.1 ng/ml to 5ng/ml, after which the proliferation of EKCs tended to be a plateau. Insulin also stimulated the proliferation of EKCs. Matched with EGF, insulin made EKCs produce proliferative synergistic effects. EGF matched with PDGF or/and TGF-β didn't raise the CPM values of EKCs, or even made them decline. PHA, at the lower concentration stimulated, but, at the higher concentration, inhibited the proliferation of EKCs, showing a typical, parabolic dose-effect curve. Stimulated by PHA and EGF, the proliferative reaction of EKCs showed some complex, but regular changes. These results suggested to us: 1. Above mentioned soluble growth factors play an important role in the proliferation of EKCs in vitro. 2. The effects of EGF upon the proliferation of EKCs may be influenced by, or due to,the participation of other growth factors, insulin and exogenours fators。
