p21^(ras)在大肠肿瘤表达的定量检测及其意义

摘　　要：应用流式细胞术定量检测10例正常大肠粘膜、27例大肠腺瘤及32例大肠腺癌的p21^(ras)及DNA含量,结果发现:①正常大肠粘膜、大肠腺瘤及大肠癌的p21^(ras)表达量依次递增,并随大肠腺瘤不典型增生程度的加重以及瘤体的增大而增多;②大肠腺瘤组p21^(ras)表达增强呈阳性者的DI值、SPF值显著高于p21^(ras)阴性者,而大肠癌组仅SPF值有显著性差异;③DNA含量呈异倍体的大肠腺瘤及腺癌的p21^(ras)表达量显著高于DNA二倍体者本研究从p21^(ras)表达的量变意义上说明:①p21^(ras)过表达促进了大肠肿瘤细胞的异常增殖,并导致细胞DNA含量异常,出现DNA异倍体;②p21^(ras)表达量的变化在大肠腺瘤向腺癌恶性转化的质变过程中可能具有重要作用。In this study p21^(ras) expression and DNA content were quantitatively detected using flow cy- tometry in 10 normal mucosas, 27 adenomas, and 32 adenocarcinomas of the colorectum. The results showed that ①quantities of p21^(ras) expression rose stepwise in normal control, adenoma and carcinoma, and rose with the increase of the degree of dysplasia and the size of tumor in adenomas; ②both DI and SPF values in the adenomas of enhanced p21^(ras) expression in the positive were significantly higher than those in the negative, and it is only true of SPF value in colorectal cancers; ③ in colorectal adenomas and cancers with DNA aneu- ploid, the quantity of p21^(ras) expression was higher when compared to diploidy tumors of colorectum. Our data indicate that ①overexpression of p21^(ras) can promote abnormal proliferation of colorectal tumor cells, leading to the abnormality of cellular DNA content and producing DNA aneuploid; ②the changes of the amount of p21^(ras) expression may play an important role in the course of qualitative alteration of colorectal adenoma to adenocarcinoma.
