摘  要:采用mRNA斑点杂交和免疫蛋白印迹方法,探讨了高温、一氧化碳(CO)以及它们联合作用对大鼠肝脏热应激蛋白70ku(HSP70)mRNA、HSP70蛋白合成的影响;使用纯化的HSP27、HSP60、HSP70、热应激同源蛋白70ku(HSC70)、HSP89α和HSP89β,调查了接触高温、CO工人血浆中相应的抗体,也调查了这些工人中血浆游离氨基酸浓度。结果:在应激处理之后,大鼠肝脏HSP70mRNA、HSP70蛋白明显增加,高温和CO联合作用对HSP70mRNA、HSP70诱导表现为协同作用,但高温起主要作用。接触高温、CO工人血浆中HSP27和HSP70抗体阳性率明显增高(P<0.05),但在CO组是最高的。尽管以前的调查结果表明这些工人摄入蛋白质不足,但他们血浆中游离氨基酸却趋向增高,特别是蛋氨酸、色氨酸,因为主要HSP70几乎不含这两种氨基酸。结果提示了这些工人所面临的主要问题是如何促进这些氨基酸的利用,减轻职业有害因素应激时正常蛋白的抑制。The effects of high temperature or carbon monoxide or both of them on rat liver's HSP70 mR-NA and HSP70 syntheses with dot hybridization of mRNA and western blot were observed. Antibodies of HSP27,HSP60,HSP70,HSC70,HSP89α and β in the workers exposed to heat or carbon monoxide or both of them using the corresponding purified antigens and their plasma free amino acids were analysed. The results showed that after heat stress treatment, HSP70 mRNA and HSP70 increased significantly and there was a synergism in the combined effects of both high temperature and carbon monoxide on the induction of HSP70 mRNA and HSP70 synthesis, with the former playing a major role. Positive rate of plasma anti-HSP27 and HSP70 in the workers working in the environment where extreme heat or carbon monoxide existed increased significantly(P<0.05), and that in carbon monoxide-exposed group was the highest. Although our previous results indicated that these workers took no enough proteins, plasma free amino acids in these workers tend-ed to increase, especially in methionine and tryptophan, because major HSP70 almost did not contain these two kinds of amino acids. These results indicated that the major problems these workers faced were how to facilitate the use of plasma free amino acids and reduce the inhibition of synthesis of normal proteins when these occupational harmful factors occurred.
