摘  要:转化生长因子-β1(TGF-β1)反义寡核苷酸能使骨髓中幼稚的CD34^+细胞从静息期进入细胞周期,从而促进CFU-GEMM及早期BFU-E的集落形成。由于干细胞因子(SCF)对CD34^+细胞的集落形成也有类似的作用,作者对SCF的作用是否与阻断TGF-β的抑制效应有关进行了研究。结果表明,与TGF-β1反义核酸、SCF单用相比,两者合用对骨髓CD34^+细胞集落形成的促进作用优于递加作用,对脐血的促进作用略低于递加作用。这说明SCF仅可使部分被TGF-β维持在静息期的脐血造血细胞进入细胞周期。当TGF-β1反义寡核苷酸和SCF合用时,脐血中CFU-GEMM的绝对数比骨髓中CD34^+细胞多4倍。这与我们曾观察到的脐血所含的CD34^+CD38^-细胞,即对TGF-β1反义寡核苷酸有反应的细胞群比骨髓中多4倍的结果相一致。It was demonstrated that TGF - (31 antisense oligodeoxynucleotides can release early CD34 + bone marrow progenitors from quiescence, and increase the numbers of mixed and large erythroid colonies. As stem celi factor (SCF) has a similar effect on colony formation by CD34+ cells, we tested whether this factor acts by blocking the inhibitory ef fects of TGF - β. That this is not generally the case was demonstrated by the finding that the combination of TGF - β1 antisense with SCF in cultures of CD34+ bone marrow celis yielded enhanced colony formation that was more than additive when com-pared to cultures containing the single agents. This combination also yielded enhanced colony formation by CD34 + umbilical cord blood cells, but in this case, the effect was slightly less than addivive. Thus in cord blood, some, but not all, of the progenitors that are maintained in quiescence by TGF-β1 can be triggered into cycle by SCF. However, the absolute number of CFU - GEMMs found in the antisense TGF - β1 plus SCF cultures of cord blood was 4 - fold higher than that found in comparable bone marrow cultures. These data correlate well with our previous observations that umbilical cord blood contains 4 fold more CD34+ CD38- cells, the population found to respond to TGF - β1 antisense oligodeoxynucleotides.
