摘 要:目的研究肺癌细胞DNA倍体类型和核仁组织区嗜银蛋白(AgNOR)及两者之间的相互关系。方法对36例肺癌石蜡标本进行细胞AgNOR定量计数和DNA含量的流式细胞术(FCM)检测。检测结果进行t检验和X2检验。结果二倍体肺癌细胞AgNOR计数(1.65±0.87)显著低于异倍体肺癌细胞者(3.09±1.03)细胞周期中S期细胞<20%的肺癌细胞AgNOR计数(1.38±1.02)显著低于S期细胞≥20%者(2.77±1.14)。在二倍体肿瘤中,分化Ⅱ级和Ⅲ级细胞的AgNOR计数显著高于分化Ⅰ级者,但在Ⅱ、Ⅲ级之间AgNOR计数则无显著差异;分化Ⅲ级肿瘤的异倍体检出率显著高于分化Ⅰ、Ⅱ级肿瘤,但在Ⅰ、Ⅱ级之间异倍体检出率则无显著差异。结论细胞DNA含量和AgNOR计数与肺癌细胞的增殖状态、恶性程度及预后有密切相关性;在病理分级中以上两种检测具有互补性。The aim of this project is to investigate the relationships between DNA content and AgNOR in lung cancer.METHODS 36 cases of paraffin-embedded lung cancer specimens were analysed by FCM for DNA contents and AgNOR count. X2 was used for statistical study.RESULTS The AgNOR counts were 3.09±1.03 in lung cancer with aneuploid DNA contents which were significantly higher than the contents obtained from lung cancer (1.65±0.87) with diploid DNA content (P<0.05). On the other hand, the cells in S phase less than 20% showed AgNOR counts lower than the amount with S phase greater than 20%. In diploid tumors, cells with gradeⅡand Ⅲ differentiation had higher AgNOR counts than gradeⅠcells, while there was no significant difference in AgNOR counts between cells with gradeⅡand Ⅲ differentiation. CONCLUSIONS There is a marked correlation between cellular DNA content and AgNOR count to proliferation status, degree of malignancy and prognosis; the two methods of examination are reciprocally helpful and might be a useful indicator for prediction in lung cancer.
