摘  要:采用大鼠腹腔一次性注射二乙基亚硝胺(DEN)作为启动剂结合部分肝切除的短期诱癌实验模型,应用图像分析法测定肝癌癌前病变内γ-谷氨酸转肽酶(γ-GT)活性的变化。实验结果发现Ⅰ组(DEN+PB)γ-GT活性高于Ⅱ组(DEN+H2O);Ⅰ组第20周γ-GT活性高于第12周,表明苯巴比妥可以诱导γ-GT活性重现,且随着实验过程延长,γ-GT活性逐渐增高。本实验说明图像分析法可以结合形态学变化在组织原位测定肝癌癌前病变内的γ-GT活性,简便、易行,优于生化法,值得进一步推广。Rats injected intraperitoneally with a single dose of diethylnitrosamine (DEN) as initiating agent in association with a partial(two-thirds)hepatectomy were used as a short-termexperimental model in this study.Image Analysis Technique was applied to determine theactivity of γ- glutamyl transpeptidase(γ-GT)enzyme in preneoplastic lesions of hepatoma.The results indicated that the γ-GT activity of group I (DEN+PB)was higher than that ofgroup Ⅱ(DEN+H2O),and the γ-GT activity of the twentieth week was higher than that ofthe twelfth week in group Ⅰ.These suggest that phenobarbital may induce the recurrence ofγ-GT activity. With the prolongation of the experiment process, the γ-GT activity wasgrandually increased.Our study proves that the γ-GT activity of preneoplastic lesions ofhepatoma can be detected in tissue in situ by theimage analysis technique in combination withthe morphological changes. Thus this technique is considered simple, convenient and applicable. It is superior to biochernical method and should be further popularized.
