The transport of metabolites to and from the chondrocytes has to take place through the matrix and hence depends on its composition and structure. A major parameter controlling the properties of cartilage is the proteoglycan (PG) content of the matrix, whose small variations will lead to considerable changes in the behavior of the tissue. It has been verified that oxygen species can damage PG and glycosaminoglycan (GAG) in different manners, but fulvic acid (FA), extracted from drinking water of Kaschin-Beck disease (KBD) frequently-occurring area, causes no direct damage. In order to investigate the effects of FA and reactive oxygen species on the pathological process, we examine the composition variations of PGs secreted by the cultured chondrocytes of chicken embryonic limb in the presence of FA and reactive oxygen species, as well as the effect of abnormal PGs on the hydroxyapative (HAP) crystal growth. Comparing these effects with the influence of 5-bromodeoxyuridine (5-BrdUrd) on the chondrocytes under the same conditions we assume that reactive oxygen species is related to chrondrocyte dedifferentiation to some extent.
