摘  要:目的 探讨 0 75mM丁酸钠对肝卵圆细胞甲胎蛋白 (AFP)表达和向细胞外分泌AFP的影响。方法从喂饲含 0 1%乙硫氨酸的胆碱缺乏性饮食 4~ 6周的大鼠肝脏中分离出肝卵圆细胞。噻唑蓝 (MTT)比色法做生长曲线 ,观察 0 75mM丁酸钠对肝卵圆细胞生长的影响。收集 0 75mM丁酸钠处理 4天后的肝卵圆细胞及其培养上清液。将细胞裂解后 ,用WesternBlot检测胞内AFP的表达水平。用酶联免疫吸附法 (ELISA)测定培养上清液中的AFP浓度。结果 生长曲线的结果显示 0 75mM丁酸钠对肝卵圆细胞的生长有一定的抑制作用。WesternBlot的结果提示0 75mM丁酸钠能显著降低肝卵圆细胞AFP的表达水平 (P <0 0 5 ) ,但ELISA法测定的培养上清液中AFP的含量却没有显著改变。结论  0 75mM丁酸钠对肝卵圆细胞的生长有一定的抑制作用 ,并能降低肝卵圆细胞AFP的表达水平。Objective Hepatic oval cells,the progenitors for liver regeneration,is one of the hot points for liver disease researches This paper was to evaluate the effects of 0 75 mM sodium butyrate on alpha-fetal protein(AFP)expression and secretion of hepatic oval cells Methods Hepatic oval cells were isolated from rats fed with choline-deficient diet supplement with 0 1%(w/w)ethonine for 6 weeks Growth curve was made by MTT methods after 0 75mM sodium butyrate treatment We collected the cultured oval cells and the supernatant after oval cells were treated by 0 75mM sodium butyrate for 4 days Then,western blot and ELISA were used to test the AFP expression in the collected cell lysate and AFP secretion in the supernatant,respectively Results 0 75 mM sodium butyrate could inhibit the growth of hepatic oval cells The AFP expression of hepatic oval cells markedly reduced after treated by 0 75mM sodium butyrate While the AFP level in supernatant didn't change significantly Conclusion 0 75 mM sodium butyrate could inhibit the growth of hepatic oval cells and could reduce AFP expression greatly
