我国不同地区多房棘球蚴原头节蛋白组分及抗原性的分折

摘　　要：本文报告了应用 SDS—PAGE 银染法对我国三个不同地区来源的多房棘球绦虫原头节蛋白组分进行了比较分析,并用三个不同来源多房棘球蚴免疫血清及泡球蚴病人血清对上述样本进行了交叉免疫印渍分析.在这些实验中同时用细粒棘球蚴,猪囊尾蚴样本做对照.5种样本在 SDS—PAGE 银染图谱上有十分明显的区别,在多房棘球蚴样本中,新疆与四川样本之间比较相似,宁夏样本在23kDa 和24kDa 处有两条特异性区带,用三个不同来源多房棘球蚴免疫血清做交叉免疫印渍分析的结果表明,同型之间反应区带较多,如宁夏样本与本身免疫血清的反应中在约31kDa～35kDa 区段产生5条免疫反应带,而新疆和四川样本各只有2条反应带,这表明有3条抗原带是宁夏样本所特有的。用一例新疆本地的泡球蚴病人血清识别三个地区样本的结果也显示出明显的差别.这些结果进一步说明了特异性和非特异性抗原表位在不同分子量肽链上的分布特点,可以做免疫分类的基础,表明我国三个地区多房棘球绦虫在蛋白成份、抗原性及抗原分布上的不同.SDS—PAGE silvered staining method was used to analyse soluble protein componants of protoscoleces of Echinococcus multilocularis from Xinjiang,Ningxia and Sichuan provinces,immunoblot analysis was used to analyse isolates from these three regions with re- spective immune serum.E.granulosus and Cysticercus cellulosae were used as controls.The SDS-PAGE silvered staining profiles of the three isolates differed greatly.E. multilocularis isolates from Xinjiang and Sichuan were relatively similar in the profiles,and those from Ningxia showed two specific zones at 23 kDa and 24kDa. Results of cross immunoblot analysis with immune sera against E.multilocularis from these three different regions showed more homotypic reaction zones,for example,the Ningxia isolates and their homotypic immune sera produced 5 immune reactive zones in the interval between 31kDa and 35kDa,whereas isolates from Xinjiang and Sichuan had only two reaction zones each,indicating that the three antigenic zones in this interval were specific for Ningxia isolates.Results of immunological recognization of isolates from the three regions by serum from an indigenous patient with alveolar hydatidosis in Xinjiang also showed significant dif- ferences in immunoblot profiles.
