摘 要:目的研究Bcl-2反义核酸对人神经母细胞瘤细胞株SK-N-SH的增殖和凋亡的作用。方法设Bcl-2反义核酸ASODN组、正义核酸SODN组、脂质体组及空白对照组。MTT法检测Bcl-2反义核酸对SK-N-SH细胞增殖的抑制作用;流式细胞术检测细胞周期及细胞凋亡情况。结果 ASODN组72h细胞增殖抑制率达(68.24±2.41)%,较空白对照组差异显著(P<0.01);ASODN可使SK-N-SH细胞周期阻滞于G2/M期,72h细胞凋亡率达(23.25±2.34)%。结论 Bcl-2反义核酸可抑制SK-N-SH细胞增殖并诱导其发生凋亡。Objective To investigate the effect of Bcl-2antisense oligonucleotides on the apoptosis and proliferation of human neuroblastoma cell lines SK-N-SH.Methods There were four groups in our experiment:Bcl-2antisense oligonucleotides group,sense oligodeoxynueleotides group,Lipofectamine group and control group.MTT assay was used to detect the inhibition ratio of different groups.Flow Cytometry Method was used to test the apoptosis and cell cycle of SK-N-SH cell.Results The proliferation of SK-N-SH cell in Bcl-2antisense oligonucleotides group was significantly inhibited compared with control group(P<0.01);Cell cycle in Bcl-2antisense oligonucleotides group was arrested in G2/M stage,and the apoptosis rate was(23.25±2.34)%.Conclusion Bcl-2antisense oligodeoxynucleotides can inhibit the SK-N-SH cell proliferation and enhance the cell apoptosis rate.
