摘 要:目的 观察姜黄素对肺癌NCI-H446细胞的体外放疗增敏作用,探讨其可能的机制.方法 体外培养肺癌NCI-H446细胞,分为对照组、姜黄素处理组、X射线干预组以及联合干预组.对照组仅给予等体积培养基,姜黄素处理组给予不同浓度姜黄素处理48 h,X射线组采用X射线照射(剂量率250 cGy/min),联合治疗组细胞先采用姜黄素处理,之后使用X射线照射.采用MTT法检测各处理组细胞增殖情况,流式细胞术检测细胞凋亡,Western blot检测细胞中p53、p21、Bax、Bcl-2蛋白表达.结果 不同浓度的姜黄素对人肺癌NCI-H446细胞生长具有抑制作用;剂量为6~12 Gy的X线处理后,也可显著抑制NCI-H446细胞增殖;不同浓度姜黄素和8GyX线处理后,NCI-H446细胞增殖进一步降低,并与姜黄素浓度呈一定的剂量依赖性.流式细胞术结果显示:姜黄素和X射线照射均可促进细胞凋亡;2者联合使用与单纯姜黄素或X射线照射相比细胞凋亡显著增加,差异均有统计学意义(P<0.05).Western blot结果显示:姜黄素与X射线均能诱导NCI-H446细胞中p53、p21和Bax蛋白表达,降低Bcl-2蛋白表达(P<0.05).X射线与姜黄素联合处理后,p53、p21和Bax以及Bcl-2蛋白含量比单独作用变化显著,差异有统计计学意义(P<0.05).结论 姜黄素对人NCI-H446细胞具有放疗增敏作用,其机制与诱导细胞凋亡,上调p53、p21蛋白及下调Bel-2/Bax表达有关.Objective To observe the radiosensitizing effect of curcumin on lung cancer NCI-H446 cell in vitro and to investigate the possible mechanism. Methods Lung cancer NCI-H446 cell were cultured in vitro and divided into four groups: control group,curcumin group,X-ray group and combined intervention group. Control group was given equal volume of medium,curcumin group was treated with 20 μmol /L curcumin for 48 h,X-ray group was irradiated by X-ray(dose rate was 250 cGy /min) and combined intervention group was treated with curcumin and X-ray. The proliferation of NCI-H446 cell in all groups were detected by MTT method,cell apoptosis were detected by flow cytometry and the protein expression of p53,p21,Bax,Bcl-2 were detected by Western blot. Results The growth of human lung cancer NCI-H446 cell were inhibited with different concentrations of curcumin and 6 ~ 12 Gy X-ray irradiation,and inhibited more in combined intervention group,in curcumin dose-dependent manner. Flow cytometry results showed that,curcumin or X-ray irradiation could all induce apoptosis,the cell apoptosis increaced significantly when treated with both(P < 0. 05). Western blot results showed that,either curcumin or X-rays could induce p53,p21 and Bax protein expression in NCI-H446 cell and decrease Bcl-2 protein expression(P < 0. 05). After the combination of X-rays and curcumin,the content of p53,p21,Bax and Bcl-2 protein was significantly changed(P <0. 05). Conclusion Curcumin has radiosensitizing effect on lung cancer NCI-H446 cell. The mechanism is connected with the inducing of apoptosis,increasing p53,p21 protein expression and decreasing the Bcl-2 /Bax ratio.
