摘 要:目的探讨血管内皮细胞生长因子(VEGF)-460 C/T和色素上皮细胞衍生因子(PEDF)Met72Thr T/C基因多态性与增殖性糖尿病视网膜病变的关系。方法运用高分辨率溶解(HRM)曲线法分析77例健康对照人群和160例糖尿病视网膜患者(包括68例增殖性糖尿病视网膜病变PDR和92例非增殖性糖尿病视网膜病变NPDR)基因型,比较其基因型和等位基因频率。结果 (1)PEDF Met72Thr T/C基因多态性和增殖性糖尿病视网膜病变的发生无相关性。(2)增殖性糖尿病视网膜病变(PDR)组VEGF-460CC基因型频率C等位基因频率明显高于NPDR组和对照组(P<0.05)。(3)等位基因C携带的基因型的PDR发病率是TT基因型的2.39倍。(4)VEGF-460C/T基因多态性以及病程是2型糖尿病发生PDR的危险因素。(5)VEGF-460C/T和PEDF Met72Thr T/C两者基因多态性对于增殖型糖尿病视网膜病变具有协同作用。结论 VEGF-460C/T基因多态性与2型糖尿病PDR发生有关,C等位基因可能是2型糖尿病PDR的易感基因。VEGF和PEDF基因型的交互作用对增殖性糖尿病视网膜病变的发生表现为协同作用。Objective To investigate the relationship between polymorphism of vascular endotnelial growth factor(VEGF)-460C/T and pigment epithelium derived factor(PEDF)Met72Thr T/C with diabetic retinopathy.Methods HRM(High Resolution Melt)was wsed to analyze the gene polymorphism of 77healthy controls and 160independent patients with type 2diabetes(68patients with proliferative diabetic retinopathy,PDR and 92patients with non-proliferative diabetic retinopatlyl,NPDR),and the genotype and allele frequencies were among different groups.Results(1)There was no significant correlations between PEDF Met72Thr and PDR.(2)The freauencies of VEGF-460CC and C allele were significantly higher in patients with PDR than those with NPDR and controls(P<0.05).(3)The odds ratio(OR)of PDR for the carriers of VEGF-460Cto the TT genotype was 2.39.(4)Logistic regression analysis revealed that the VEGF-460 C/T polymorphism and duration were independent risk factors of PDR.(5)These results demonstrated that the VEGF gene,in cooperation with the PEDF gene,may contribute to the development of DR.Conclusions VEGF-460C/T genetic polymorphism was associated with PDR.C allele is probably a susceptible gene of PDR.These results demonstrated that the VEGF gene,in cooperation with the PEDF gene,may contribute to the development of DR.
