摘　　要：目的研究CD137L在非小细胞肺癌中的表达,及其对细胞增殖、迁移和侵袭的影响,探讨其可能的分子生物学机制。方法通过体外构建CD137L高表达慢病毒载体[A549-TC-1(+)]及CD137L-RNA干涉慢病毒载体[A549-TC-1(-)]和空白载体(A549-control),分别转染肺癌A549细胞。采用Western blot检测3组细胞CD137L蛋白的表达;MTT法检测细胞增殖情况;Matrigel transwell小室侵袭实验研究肺癌细胞侵袭能力;ELISA方法检测细胞上清IL-6和IL-8的水平。结果 Western blot结果显示:与A549细胞相比,CD137L蛋白在A549-TC-1(+)细胞中表达明显增多,A549-TC-1(-)细胞中表达明显减少,差异有统计学意义(P<0.05),在转染A549-control中无明显变化,表明3种病毒载体可用于后续实验。MTT细胞增殖实验及Matrigel Transwell小室侵袭实验结果显示:与A549-control相比,A549-TC-1(+)细胞增殖、迁移及侵袭能力均明显增强,A549-TC-1(-)细胞增殖、迁移及侵袭能力均明显减弱,差异均有统计学意义(P<0.05)。ELISA检测结果显示:A549-TC-1(+)细胞中细胞因子IL-6、IL-8的水平明显高于A549-TC-1(-)细胞组和正常A549细胞组。结论 CD137L对非小细胞癌细胞的增殖、侵袭具有正性调节功能,其作用机制可能与促进IL-6、IL-8的分泌进而增强肿瘤的生长、增殖和侵袭能力有关。Objective To study CD137 L expression in non-small cell lung cancer,its effect on cell proliferation,migration and invasion and explore the molecular mechanisms of non-small cell lung cancer invasion and metastasis. Methods Lentiviral vectors of CD137 L overexpression,CD137L-RNA interference and A549 control were constructed and transfected into A549 cell. CD137 L protein expression in three groups were detected by Western blot; cell proliferation assay were conducted by MTT; cancer cells proliferation,invasiveness were measured by Matrigel transwell chamber invasion assay; IL-6 and IL-8 levels in cell supernatants were detected by ELISA method. Results Western blot results showed that: compared with A549 cell,CD137 L protein expression in A549-TC-1( +) cell increased significantly,A549-TC-1(-) cell decreased significantly,the differences were all statistically significant( P < 0. 05),and there was little change in transfected A549-control cell,which indicating that three kinds of viral vectors can be used for subsequent experiments.. MTT and matrigel transwell chamber invasion assay results showed that: compared with A549-control cell,A549-TC-1( +) cell proliferation migration and invasion capacity were enhanced,and A549-TC-1( +) cell were weakened,the differences were all statistically significant( P < 0. 05). ELISA results showed that cell serum IL-6,IL-8 levels in A549-TC-1( +) cell were all significantly higher than A549-TC-1(-)cell and normal A549 cell a groups. Conclusion CD137 L can regulate non-small cancer cell proliferation and invasion positivily,the mechanism may related to increasing IL-6,IL-8 secretion and thereby enhancing tumor growth,proliferation and invasion.
