摘  要:为探讨基质金属蛋白酶-1(MMP-1)、基质金属蛋白酶-9(MMP-9)和集硅酶-1在低聚果糖诱导的马急性蹄叶炎中发挥的作用,将24匹蒙古马随机分为试验组和对照组,每组12匹。试验组灌服低聚果糖,从灌服后第0到72小时,每隔4h采集1次颈静脉血(5mL),3 000r/min离心10min后用ELISA检测血浆中MMP-1、MMP-9和集硅酶-1的质量浓度。结果显示,MMP-1、MMP-9和集硅酶-1的质量浓度分别在灌服后第24、28和48小时达到峰值,峰值分别为37.96ng/mL±4.91ng/mL、199.37ng/mL±25.39ng/mL和15.11ng/mL±1.50ng/mL,与对照组差异性显著(P<0.05)。结果表明,MMP-1、MMP-9和集硅酶-1的质量浓度随时间发生较大变化,可能在急性蹄叶炎的发展过程中起到重要作用,其中MMP-9可能是关键的分子指标。To explore effects of metalloproteinase-1(MMP-1),metalloproteinase-9(MMP-9)and aggrecanase-1in oligofructose-induced equine acute laminitis,24 Mongolian horses were divided into experimental and control groups randomly.The horses in the experimental group were fed oligofructose.Jugular venous blood samples were collected once every 4hours from hour 0to 72post-treatment,and were centrifuged for 10 minutes at 3 000r/min.The concentrations of MMP-1,MMP-9and aggrecanase-1were detected by ELISA.In result,MMP-1,MMP-9and aggrecanase-1were reached a peak respectively at 24h(37.96ng/mL±4.91ng/mL),28h(199.37ng/mL±25.39ng/mL)and 48h(15.11ng/mL±1.50ng/mL)posttreatment,indicating that the difference was significant compared with the control group(P<0.05).The results demonstrated that the concentrations of MMP-1,MMP-9and aggrecanase-1changed greatly as time goes,and MMP-1,MMP-9and aggrecanase-1play important roles in the development of acute laminitis,but MMP-9may be a key indicator.
