摘 要:目的:探讨神经源性分化因子NeuroD在大鼠急性脊髓损伤过程中的表达及意义.方法:以改良Allen''s法建立急性脊髓损伤(SCI)大鼠模型,以假手术组为对照,应用免疫组织化学、半定量RT-PCR、免疫印迹分别检测脊髓损伤后不同时间点NeuroD的表达变化.结果:NeuroD特异性表达于脊髓灰质,与对照组相比,急性脊髓损伤后3d,NeuroD mRNA的相对表达量明显增加,5d达到峰值,7d后回落;Neurod的蛋白相对表达量则在急性脊髓损伤5d后明显升高,持续至第7天,14d明显回落.结论:急性脊髓损伤可上调NeuroD表达水平,提示NeuroD参与了急性脊髓损伤后的病理过程.Objective:To investigate the expression and significance of neurogenic differentiation factor NeuroD in the process of acute spinal cord injury.Methods:Modified Allen’s method was used to establish rat models of acute spinal cord injury(SCI).The expression of NeuroD in sham-operated group and SCI group were detected by immunohistochemistry,semiquantitative RT-PCR and Western blotting respectively at different time points.Results:NeuroD specifically expressed in the spinal cord gray matter;Compared with the control group,the relative expression level of the neuroD mRNA in SCI group increased significantly at 3rd day after spinal cord injury,and reached the peak at 5th day,and then declined at 7th day.However,the relative expression level of proteins was significantly elevated at 5th day after spinal cord injury and lasted to 7th day,and then fell obviously at 14 th day.Conclusion:Acute spinal cord injury upregulates the NeuroD expression levels,which suggest NeuroD maybe involved in the pathogenesis of acute spinal cord injury.
