纳洛酚对脑缺血大鼠皮层体感区Ca^(2+)-ATP酶活性的影响

摘  要:目的 观察脑室注射纳洛酚对脑缺血大鼠大脑皮层体感区Ca2 + ATP酶活性的影响 ,探讨纳洛酚对急性大鼠脑缺血性损伤的保护机制。方法 建立大鼠大脑中动脉栓塞模型 ,给予生理盐水、药物后检测Wistar大鼠皮层体感区组织匀浆上清液Ca2 + ATP酶的活性。观察正常大鼠脑室注射 10 μl生理盐水与大脑中动脉栓塞后脑室分别注射生理盐水 10 μl,纳洛酚 1μg/ 10 μl、4 μg/ 10 μl、8μg/ 10 μl及纳洛酮 8μg/ 10 μl对Ca2 + ATP酶活性的影响。结果 脑缺血组与生理盐水对照组相比Ca2 + ATP酶活性显著降低 (P <0 .0 1) ,给予纳洛酚与纳洛酮的实验组与缺血组相比 ,Ca2 + ATP酶活性升高 (P <0 .0 1) ,而且 1μg/ 10 μl,4 μg/ 10 μl和 8μg/ 10 μl纳洛酚 3组呈现随剂量增加酶活性也随之提高的量效关系 (P <0 .0 1) ,4 μg/ 10 μl和 8μg/ 10 μl纳洛酚提高酶活性的作用优于纳洛酮 (P <0 .0 1)。 结论 纳洛酚可能通过拮抗阿片受体提高大鼠急性大脑中动脉缺血后皮层体感区Ca2 + ATP酶活性 。Objective To study the effects of intracerebrovertricular injection of nalorphine on the Ca 2+ ATPase activity in cerebral ischemia rats and discuss the protective mechanism of nalorphine on acute ischemic injury rats.Methods The intracerebroventricular injection of saline and drugs were used in the middle cerebral artery occlusion model of Wistar rats,Ca 2+ ATPase of brain tissues was measured.To investigate the effects of intracerebroventricular injection of 10μl saline,nalorphine (1μg/10μl,4μg/10μl and 8μg/10μl) and 8μg/10μl naloxone on Ca 2+ ATPase activity of somatosensory area of rat cerebral cortex.Results Compared cerebral ischemia group with control group,it demonstrated that cerebral ischemia injuration could decrease Ca 2+ ATPase activity significantly(P<0.01).Intracerebroventricular injection of nalorphine and naloxone could increase the Ca 2+ ATPase activity(P<0.01),with the increase of nalorphine doses,the activity also increase in the 1μg/10μl,4μg/10μl and 8μg/10μl nalorphine groups respectively,the influence of 8μg/10μl nalorphine group was better than 8μg/10μl naloxone group(P<0.01).Conclusion These results indicate that nalorphine can increase the activity of Ca 2+ ATPase after the middle cerebral ischemia by blocking the morphine receptor in CNS,so nalorphine has the protective influence on cerebral ischemia rats.
