摘　　要：目的 :观察PDGF B链基因三链形成寡核苷酸 (triplex formingoligonucleotide,TFO)对C6胶质瘤细胞PDGF B链基因表达和细胞生长的作用。探索PDGF B链基因TFO作为抗肿瘤治疗新药的可能性。方法 :应用MTT法观察PDGF B链基因TFO对C6胶质瘤细胞生长的抑制作用 ;应用流式细胞技术观察PDGF B链基因TFO对C6胶质瘤细胞PDGF B链基因表达的影响。结果 :PDGF B链基因TFO对C6胶质瘤细胞PDGF B链基因的表达和细胞生长有明显抑制作用 ,而且抑制作用存在浓度依赖性。结论 :PDGF B链基因TFO能够抑制C6胶质瘤细胞PDGF B链基因的表达和细胞生长 ,有望成为抑制PDGFObjective:To clarify the inhibition of the expression of PDGF-B chain and the proliferation f C_6 glioma cells by triplex-forming oligonucleotide(TFO). Methods:C_6 glioma cell growth was assessed by microculture tetrazolium dye (MTT) assay two days after adding different dosage of triplex-forming oligonucleotide. PDGF-B chain expression was quantified by immunofluorescence flow cytometric analysis. Results:triplex-forming oligonucleotide can obviously inhibit the PDGF-B chain expression and the proliferation of C_6 glioma cells in concentration-dependent fashion. Conclusion:Triplex-forming oligonucleotide can effectively suppress the C_6 glioma cell PDGF-B chain expression and the cell proliferation.
