散发性乳腺癌中BRCA1和cyclinB1的表达

摘　　要：目的　探讨BRCA1和cyclinB1在散发性乳腺癌的表达及意义。方法　应用免疫组化S P法检测 2 0例乳腺腺病和 5 2例散发性乳腺癌中BRCA1和cyclinB1的表达。结果　所有乳腺腺病中BRCA1均呈阳性表达 ,散发性乳腺癌中BRCA1表达率和强度均下降 (P <0 0 1) ,BRCA1表达下降或丧失与散发性乳腺癌组织学分级有关 (P <0 0 5 )。cyclinB1在乳腺腺病和散发性乳腺癌中表达阳性率分别为 35 0 % (7/ 2 0 )和 6 9 2 % (36 / 5 2 ) ,差别有统计学意义 (P <0 0 1) ,cyclinB1过表达与组织学分级和肿块大小有关 (分别P <0 0 1,P <0 0 5 )。在散发性乳腺癌中BRCA1与cyclinB1的表达呈负相关 (P <0 0 1)。Purpose To investigate the expression of BRCA1 and cyclinB1 in sporadic breast carcinoma and explore the relationship between BRCA1 and cyclinB1. Methods Immunohistochemical method(S-P) was used to examine the expression of BRCA1 and cyclinB1 proteins in paraffin-embedded specimens of 20 cases of benign epithelial proliferations and 52 cases of sporadic breast carcinomas. Results All benign epithelial proliferations expressed BRCA1, while sporadic breast carcinoma expressed lower level than proliferative breast disease(P<0.01). Loss of BRCA1 expression was closely correlated with histologic grades of sporadic breast carcinoma (P<0.05). The positive rate of cyclinB1 expression in sporadic breast carcinoma was higher than that in benign epithelial proliferations (P<0.05), and the expression of cyclinB1 was closely correlated with histological grade and tumor size(P<0.01, P<0.05 respectively). There was a negative correlation between BRCA1 and cyclinB1 expression in sporadic breast carcinoma (P<0.01). Conclusions A reduced expression of BRCA1 and an over-expressed cyclinB1 may play an important role in the carcinogenesis and progression of sporadic breast carcinoma.
