肺癌SEMA3B基因的表达及其临床意义

摘　　要：目的　探讨新型候选抑癌基因SEMA3B在肺癌组织中的表达及其与肺癌发生、发展的关系。方法　采用逆转录 聚合酶链反应 (RT PCR)方法检测 5 2例肺癌组织及远癌正常肺组织SEMA3B基因产物的表达水平。结果　肺癌组织中SEMA3B的表达率为 5 1.9% (2 7/5 2 )明显低于远癌正常肺组织 10 0 .0 % (5 2 /5 2 ) ,P <0 .0 1。淋巴结转移组的表达缺失率 64 .3 % (18/2 4)显著高于未转移组 2 9.2 % (7/2 4) ,TNMⅢ期表达缺失率 72 .7% (16/2 2 )高于Ⅰ、Ⅱ期 2 5 .0 % (4 /16)、3 5 .7%(5 /14 ) ,P <0 .0 5 ;表达异常与肺癌癌瘤类型、大小、部位、患者的年龄、性别及吸烟与否均无明显关系 (P >0 .0 5 )。结论　SEMA3B表达异常可能在中国人群肺癌发生。Objective Oncogenesis and development of lung cancer.Methods The expression of SEMA3B was detected in 52 human lung cancer tissues and paired 52 non cancer tissues by To explore the relationship between SEMA3B gene expression and onc RT PCR.Results The SEMA3B transcript was identified in all non cancer tissues 100.0%(52/52) but was found only in 51.9%(27/52) carcinoma tissues ( P <0.001).Loss of SEMA3B expression was significantly high in patients with positive lymph node metastasis 64.3% (18/24) compared with those without lymph node metastasis 29.2%(7/24), P <0.05.The rate of loss of SEMA3B?mRNA expression was correlated with TNM stage ( P < 0.05), when stage Ⅲ 72.7%(16/22) is compared with stage I and II,respectively 25.0%(4/16) and 35.7%(5/14) ( P <0.05).No significant association of abnormal SEMA3B expression with histological type,differentiation grade of tumors or age,sex,and smoking index of the patients was observed ( P > 0.05). Conclusions SEMA3B is one of the new candidates of tumor suppression genes.Loss or abnormal down regulation of SEMA3B is a frequent event in lung oncogenesis,which might play an important role in the malignant progression and prognosis of lung carcinomas.
