救脑益智胶囊对D-半乳糖老化小鼠海马神经元Aβ及其相关蛋白表达的影响

摘　　要：目的　观察中药救脑益智胶囊对 D-半乳糖老化小鼠 (DGAM)海马神经元β-淀粉样肽 (β-amyloid,Aβ)及其相关蛋白表达的影响。方法　采用免疫组织化学染色和免疫印记方法 ,检测 DGAM模型海马神经元 Aβ、β-分泌酶、内质网 Aβ结合蛋白和早老蛋白 -1的表达水平 ,并观察中药救脑益智胶囊对 DGAM的保护作用。结果　对照组小鼠海马 CA1区神经元有少量 Aβ及其相关蛋白表达 ,而 DGAM海马 CA1区神经元 Aβ及其相关蛋白表达水平明显增加 ,与对照组相比差异有显著性 (P <0 .0 1 ) ,中药组上述各种蛋白的表达与对照组相比差异无显著性。结论　 DGAM海马神经元存在 Aβ及其相关蛋白表达的上调 ,中药救脑益智胶囊可明显改善模型动物中上述蛋白的异常表达 ,对神经元具有一定的营养和保护作用。Objective To examine the effect of Chinese medicine Jiunao Yizhi capsule on the changes of β-amyloid (Aβ) and related protein of D-galactose induced aging of murine brain (DGAM). Methods Immunohistochemical staining and western blotting for Aβ, beta-site APP-cleaving enzyme (BACE), endoplasmic reticulum amyloid β-peptide binding protein (ERAB) and presenilin-1 (PS-1) of DGAM were performed, the protective effect of Jiunao Yizhi capsule was observed. Results The expression level of Aβ and related proteins in the hippocampal of DGAM increased obviously as compared with control group ( P <0.01), while the result of Chinese medicine group was the same as that in normal mice. Conclusions The expression of Aβ and related protein in the hippocampal of DGAM are increased, while Chinese medicine Jiunao Yizhi capsule normalizes the expression and protects neurons.
