摘 要:镉是一种致癌物质,其致癌作用与Cd2+有着直接或间接的作用.用一定浓度的CdCl2溶液处理质粒pUC119和pCPP9,通过琼脂糖凝胶电泳分析,研究了镉对质粒DNA结构的影响.结果表明,CdCl2溶液2.5μg/mL作用12h或20μg/mL作用1h后,能导致质粒pCPP9的DNA损伤;浓度40μg/mL以上的CdCl2作用12h后,引起质粒pUC119的DNA损伤;且随着CdCl2浓度增大和作用时间的延长,对质粒DNA的损伤作用增强.Cadmium is a potent carcinogen.Interaction of cadmium with DNA was studied in order to understanding the mechanism of cadmium-induced carcinogenicity.The results showed that Cd^(2+)could react with plasmid DNA and caused DNA damage.The results also indicated that the treatments of CdCl_2 at concentration of 2.5 μg/ml for 12 h or at concentration of 20 μg/ml for 1 h caused a significant increase in the intensity of the oc DNA band as compared with the observed in the untreated pCPP9,while the treatment of CdCl_2 at above 40 μg/ml for 12 h caused damage to pUC119.The damage to plasmid DNA enhanced with the increasing of treatment concentrations and the exposure time prolonged.
