摘　　要：逆转录 HSV-TK 载体用于肝癌的转基因治疗已在动物实验证明有良好效果,但目前尚无满意的靶向表达载体可供使用,本研究采用 PCR、Southern blot 及 Northern blot 等方法,对本室构建的逆转录载体 LN.ALBTRS.TK 在体外对多种靶细胞进行转染和表达的检测,结果表明:LN.ALBTRS.TK 能转染多种肿瘤细胞,包括肝癌细胞及非肝癌细胞,并在表达白蛋白的肝癌细胞中可检测到 mRNA 的转录和HSV-TK 的表达产物,而在不表达白蛋白的非肝癌细胞中元 HSV-TK 的 mRNA 转录和 HSV-TK 的蛋白产物表达。这一结果表明:LN.ALB-TRS.TK 逆转录载体能正确的在期望的细胞中靶向表达,具备一定的组织特异性。To study the targeting expression of retroviral vector LN.ALBTRS.TK which constructed by us,we assay the expression of the hsv-tk gene production in transductive target cell lines which includ- ing hepatocellular carcinoma cell lines and non-hepatocellular carcinoma cell lines by means of PCR,South- ern blot and Northern blot.The results demonstrate that LN.ALBTRS.TK can transduct some kind of tu- mor cell lines including hepatocellular carcinoma cell lines which expressing human album protein and non- hepatocellular carcinoma cell lines which no expressing human album protein.In these transductive cell lines,the hsv-tk production expression only measured in the hepatocellular carcinoma lines which express- ing human album protein.This results syowed retroviral vector LN.ALBTRS.TK can express correctly in the target cells.
