摘  要:目的 探讨大鼠全脑照射后早期大脑皮质少突胶质前体细胞的放射性反应及MK - 80 1和NBQX对其保护作用。方法 以 1 0Gy的剂量对SD大鼠单次全脑照射后即刻分别向其大脑皮质定向注射 5mmol/L的MK - 80 1和 5 0mmol/L的NBQX各 1 μl,然后用免疫荧光组织化学法分别检测各时间点大脑皮质MyT1阳性细胞数量。结果 和假照射组相比 ,照射组大鼠照射后 7d和 1 4d时大脑皮质MyT1阳性细胞数显著增加 (P <0 .0 1 ) ;MK - 80 1组大鼠照射后各时间点大脑皮质MyT1阳性细胞数与照射组大鼠无明显差异 (P >0 .0 5 ) ;而NBQX组大鼠照射后 1d、7d和 1 4d时的MyT1阳性细胞数明显多于照射组大鼠 (P<0 .0 5 )。结论 大鼠全脑照射后早期大脑少突胶质前体细胞反应性增多 ,并有时程性变化 ;Objective To explore the early phase radio-reaction of oligedendrocytes in the cortex of rats after whole brain irradiation and the protections of MK-801 and NBQX to them. Methods 1 microliter MK-801 (5mmol/L) and NBQX (50mmol/L) are respectively injected to the cortex of SD rats under stereo-directional instrument after whole brain irradiation at dose of 10 Gy. Then we detected MyT1-positive cells in the cortex at each time-point by immunofluorescence histochemistry. Results The number of MyT1-positive cells within the cortex of rats in radiation group increases significantly at 7th and 14th day after irradiation comparing with sham radiation group (P<0.01). There is no obvious difference in the numbers of MyT1-positive cells in the cortex between the rats of MK-801 group and radiation group (P>0.05). The numbers of MyT-positive cells in the cortex of rats in NBQX group have significant increased at 1, 7th and 14th day comparing with that of rats in radiation group (P< 0.05). Conclusions The number of oligodendrocytes precursor cells in the brain of rats increases reactively in the early phase after whole brain irradiation and has alternated with observing time- points. And NBQX probably has some neuroprotective effect to oligodendrocyte precursor cells radiation injury in the early phase.
