摘  要:目的 探讨蛋白酪氨酸磷酸酶SHP 2的表达及其功能在γ射线体外诱发的白血病小鼠胸腺细胞中的变化情况 ,为进一步从细胞信号转导途径入手研究辐射致癌的分子机制奠定基础。方法 BALB c小鼠 3 3 6只分为癌变组、未癌变组及对照组 ,应用Westernblot、免疫沉淀法及生物化学方法检测各组SHP 2蛋白质的表达、酪氨酸磷酸化水平及酶活性。结果 癌变组SHP 2蛋白的表达、酪氨酸磷酸化水平及酶催化活性间存在明显相关性 ,均明显高于对照组及未癌变组 (P<0 0 1)。结论 在γ射线诱发的白血病小鼠胸腺细胞中 ,SHP 2蛋白的表达及其功能均明显增强 。Objective To investigate the expression and function of SH2 domain containing protein tyrosine phosphatase SHP 2 of thymocytes in γ ray irradiation induced leukemia in mice. Methods BALB/c mice were divided into canceration group, non canceration group, and control group. The expression, level of tyrosine phosphorylation, and enzyme catalytic activity of SHP 2 in thymocytes were detected by Western blotting, immunoprecipitation, and biochemical method. Results There was obvious correlation among the expression, level of tyrosine phosphorylation, and enzyme catalytic activity of SHP 2, which were significantly higher in the canceration group than those in the non canceration group and the control group ( P <0.01). Conclusion Enhanced expression and function of SHP 2 protein of thymocytes are found in γ ray irradiation induced leukemia in mice, which might be positively correlated with the occurrence of γ ray irradiation induced leukemia in mice.
