摘 要:目的:观察中药补阳还五汤对大鼠局灶性脑缺血后内源性神经干细胞(neuralstemcells,NSC)反应的影响探讨其可能机制。方法:84只雄性SD大鼠随机分为假手术组、模型组、补阳还五汤组,采用线栓法复制大鼠脑缺血模型,分别给予不同处理,于术后12h,1,3,5,7d处死动物,运用免疫组织化学方法检测脑缺血后各组NSC的激活。另取5只大鼠为正常对照组。结果:正常大鼠脑组织中存在少量NSC,主要分布在皮质区;脑缺血后,NSC增殖,以皮质、缺血周围区增加明显,健侧海马区可见NSC激活;1d开始增多,5d达高峰,模型组1,3,5,7dNSC数分别为(10.3±1.3),(14.2±1.7),(18.3±1.2),(16.4±1.6)个,补阳还五汤则分别为(10.8±1.1),(15.4±1.4),(23.5±1.1),(22.0±1.4)个,两组比较NSC数在5,7d时间点差异有非常显著性意义(P<0.01)。结论:局灶性脑缺血后,NSC激活、增殖,补阳还五汤能促进NSC的增殖。AIM:To observe the effect of buyang huanwu tang on the reaction of endogenous neural stem cells(NSCs)after focal cerebral ischemia in rats, so as to explore i ts possible mechanism. METHODS:Totally 84 male SD rats were randomly divided into 3 groups:sham opera tion group,model group and buyang huanwu tang group,focal cerebral ischemic mode ls were induced by the thread occlusion method,which received different treatmen ts,and then the rats were killed at 12 hours,1,3,5 and 7 days after operation.Th e activation of NSC was determined in all groups with immunohistochemistry.Anoth er 5 rats were taken as the normal control group. RESULTS:A few NSCs were observed mainly at the cerebral cortex in the normal r ats,and the number of NSCs was increased after ischemia,especially in the cerebr al cortex and the region around ischemia. The activation of NSCs was observed in the hippocampal area of the uninjured side;the number of NSCs began to increase on the 1st day,and reached its peak on the 5th day.The number of NSCs at 1st,3r d, 5th and 7th days in the model group was 10.3±1.3,14.2±1.7,18.3±1.2 and 16. 4±1.6,respectively, and that in the buyang huanwu tang group was 10.8±1.1, 15. 4±1.4, 23.5±1.1 and 22.0±1.4,respectively, and the differences of NSC number between the two groups at the 5th and 7th days were significant(P< 0.01). CONCLUSION:NSC is activated and proliferated after focal cerebral ischemia,and buyang huanwu tang can improve the proliferation of NSCs.
