摘 要:目的 :进行临床前B→O血型改造的实验动物研究 ,观察α 半乳糖苷酶对长臂猿B抗原的酶解效果及α 半乳糖苷酶酶解后的红细胞进行异种血型动物输血的安全性。方法 :用 10 0U/ml基因重组咖啡豆α 半乳糖苷酶体外处理B型长臂猿红细胞 ,使其转变为O型 ,再回输给B、A、AB型长臂猿。结果与结论 :α 半乳糖苷酶可以清除B型长臂猿红细胞表面B抗原 ,而不影响红细胞结构、功能及其在受体猿体内存活 ;α 半乳糖苷酶酶解的B型红细胞输给B、A和AB型血的长臂猿 ,未发生输血反应 ;受血猿的血液及尿液常规指标与输血前相比 ,无明显变化。Objective: To observe the effects of enzymatic treatment with α-galactosidase on B antigens on the surface of gibbon red blood cell (gRBC) and to study the safety of transfusion of enzyme-treated gRBC in animal transfusion experiment before entering clinical trial of B→O blood conversion. Methods: Gibbon red blood cells of type B were digested by 100U/ml recombinant coffee bean α-galactosidase, which resulted in B→O blood conversion. Then, the converted red blood cells were transfused to the recipient gibbons with B,A and AB blood types, respectively. Results and Conclusion: The results showed that α-galactosidase could remove blood antigen B on the surface of gibbon red blood cells efficiently. There were no harmful effects on donor red blood cell structure and function as well as on donor cell survival in recipient gibbon after α-galactosidase digestion. The results also demonstrated that no transfusion rejection happened in recipient gibbon with different blood groups. The routine blood and urine examinations of recipient gibbon were normal after enzymatically converted blood cell transfusion.
