摘 要:目的 探讨 5 氮 2′ 脱氧胞嘧啶 (DAC)对肺腺癌A5 4 9细胞株TRAG 3肿瘤抗原表达的影响。方法 采用逆转录 聚合酶链式反应 (RT PCR)及免疫组化的方法。结果 RT PCR和免疫组化方法均显示 :正常情况下 ,肺腺癌A5 4 9细胞内不表达肿瘤相关抗原TRAG 3;5 氮 2′ 脱氧胞嘧啶诱导 2 4h后 ,A5 4 9细胞内即可检测到TRAG 3mRNA和蛋白表达 ,直至检测的第 7天仍然有TRAG 3表达 ;TRAG 3蛋白表达位于细胞胞浆内。结论 5 氮 2′ 脱氧胞嘧啶可用于诱导肺癌细胞表达TRAG 3。Objective To explore the effect of 5 Aza 2′ deoxycytidine(DAC) on the expression of TRAG 3(taxol resistance associated gene 3) in A549 lung adenocarcinoma cell line.Methods Reverse transcription polymerase chain reaction(RT PCR) and immunohistochemistry(IHC) were used to detect the expression of TRAG 3 on the level of mRNA and protein.Results Both RT PCR and IHC showed that there was no TRAG 3 expression in untreated A549 cells;and expression of TRAG 3 could be detected on the level of mRNA and protein after exposure to DAC at time of 24h,3d,5d and 7d.Moreover,TRAG 3 expression was located at the cytoplasm in A549 cell line.Conclusion Our results suggest that DAC treatment may be a novel strategy to induce TRAG 3 expression,and might be used in enhancing the immunogenecity of lung adenocarcinoma.
