摘  要:目的 了解正常及视神经损伤大鼠闪光视觉诱发电位 (F VEP)及眼内应用腺病毒转染脑源性神经营养因子 (brain derivedneurotrophicfactor,BDNF)基因对F VEP的影响。方法 应用多功能电生理检查仪检测正常大鼠F VEP。建立大鼠视神经不全损伤模型 ,同时将含有BDNF基因的重组腺病毒经玻璃体腔注射到大鼠眼内 ,观察伤后 2h ,1、2、3、4周F VEP中P1波振幅和峰潜时的变化。结果 本实验系统测得的正常大鼠F VEP中P1波振幅为 (15 .5 1± 3.6 7) μV ,峰潜时为 (86 .2 0± 4 .30 )ms(n =2 0 )。视神经不全损伤后急性期P1波振幅降低 ,峰潜时延长 (P <0 .0 1)。BDNF腺病毒组P1波较对照组波幅高 ,峰潜时短 ,且在 1~ 3周具有显著性差异 (P <0 .0 5 )。结论 腺病毒介导BDNF基因转染对大鼠视神经损伤后早期功能的恢复有促进作用。Objective To study the changes of murine electrophysiology of vision after optic nerve injury and the influence to it by adenovirally delivered brain-derived neurotrophic factor(BDNF) to ocular tissues. Methods The F-VEP of normal rats was monitored with visual electrophysiology testing instrument. Adenovirus with BDNF gene was injected into vitreous of the injured eye of optic nerve crushed-model rats. The F-VEP was reexamined 2 hours later and at the 1st,2nd,3rd and 4th week after crush. The P_1 wave amplitude and latency were observed.Results The normal SD rats P_1 wave amplitude and latency were (15.51±3.67)μV and (86.20±4.30)ms(n=20) based on our experimental system. Optic nerve postinjuryed at 2 hours,the P_1 wave amplitude decreased and latency increased(P<0.01). The P_1 wave amplitude of BDNF group were higher than the control group's and the latency of BDNF group were shorter meantime. There were significent differences at the 1st ,2nd and 3rd week(P<0.05). Conclusion Adenovirally delivered BDNF to murine ocular tissues after optic nerve injury improves electrophysiology of vision during earlier period.
