摘  要:目的 研究NO是否参与脑缺血后海马神经元再生。方法 血管内栓线法阻塞大脑中动脉 1.5h后再灌注 ,制备局灶脑缺血再灌性模型。脑缺血后 1h海马内输入 10 μLNO供体硝普钠 (0 .1mmol·L- 1)和维生素C(0 .4mmol·L- 1)。于脑缺血后 12 0h ,5 溴 2′ 脱氧尿嘧啶核苷 (BrdU)掺入法测定海马齿状回的细胞扩增。于再灌注后 2 4h测定海马诱导型一氧化氮合酶 (iNOS)活性及NO水平。结果在再灌注后d 8缺血侧海马齿状回的BrdU阳性细胞数比对照侧多大约 5倍 ,硝普钠和维生素C海马内输入显著增加脑缺血诱发的海马齿状回神经细胞扩增。AIM To determine whether NO is involved in neurogenesis in dentate gyrus after cerebral ischemia. METHODS The middle cerebral artery in rats was occluded for 1.5 h by an intraluminal filament and then reperfused to prepare transient focal cerebral ischemia. Normal saline 10 μL containing 0.1 mmol·L -1 sodium nitroprusside(SNP) and 0.4 mmol·L -1 vitamin C (V C) was infused into ipsilateral hippocampus 1 h after cerebral ischemia. A bromodeoxyuridine (BrdU) labeling method was used to identify the postproliferation cells in dentate gyrus 120 h after ischemia. The measurement of inducible nitric oxide synthase activity and NO concentration in the hippocampus were performed at 24 h after reperfusion. RESULTS The density of BrdU positive cells in the ipsilateral dentate gyrus increased about 5 fold 8 d after ischemia, compared with the contralateral one. SNP and V C infusion significantly increased ischemia induced cells proliferation in the dentate gyrus. CONCLUSION NO takes place an important effect in ischemia stimulated cell reproducing in the dentate gyrus of adult rats.
