摘 要:目的 :为探讨脑缺血预处理对全脑缺血大鼠脑组织超微结构及细胞凋亡的影响。方法 :本文以 Wistar大鼠为受试对象 ,筛选后 14 4只大鼠随机分为正常对照组 (2 4只 )、假手术组 (2 4只 )、脑缺血预处理对照组 (32只 )、脑缺血组 (32只 ) ,脑缺血预处理组 (32只 ) 5组和术后 12、2 4、4 8、72 h四个时间点。采用“4 -动脉阻断”方法建立大鼠全脑缺血模型 ,脑缺血后大脑皮层、海马 CA1 区 HE染色观察组织形态学变化、电镜观察组织超微结构 ,原位末端标记 (TUNEL 染色 )法检测神经元凋亡。结果 :正常组、假手术组、脑缺血预处理对照组大脑皮层、海马 CA1 区无形态学改变 ,未见有细胞凋亡。脑缺血组大脑皮层、海马 CA1区神经元缺失明显 ,12 h、2 4 h、4 8h、72 h神经元凋亡逐渐加重。与脑缺血组相比 ,脑缺血预处理组大脑皮层、海马 CA1 区神经元损伤小 ,水肿程度轻 ,神经元形态恢复快 ,凋亡细胞数目少。结论 :脑缺血预处理对全脑缺血大鼠大脑皮层、海马 CA1 区神经元具有保护作用 ,可以抑制全脑缺血大鼠大脑皮层、海马 CA1 区神经元凋亡。Objective: To explore whether preacupuncture can induce the cerebral tolerance to ischemia and has a protective effect on the brain and an influence upon neuronal apoptosis. Methods: We divided 144 rats into five groups at random:normal control group(24),sham-operation group(24),ischemia pretreatment control group(32),ischemia group(32)and ischemia pretreatment group(32)and we set the time at 12h,24h,48h and 72h after operation.Acupuncture was used before brain ischemia in four-vessel occlusion (4-V0) model rats. The HE method was used to observe histomorphologucal changes. The TUNEL method and electron microscope were used to observe neuron apoptosis of the cerebral cortex and hippocampal CA 1 section. Results:There were no histomorpological changes and no neuronal apoptosis in the cerebral cortex and hippocampal CA 1 section of the sham-operation group,normal control group and ischemic pretreatment control group.The neuron damage was evidence in the cerebral cortex and hippocampal CA 1 section of the ischemia group,and 12,24,48 and 72h after operation the damage became serious.Compared with ischemia group,the ischemia pretreatment control group showed less neural damage and dropsy,rapid recovery of neuron form and fewer apoptostic cells in the cerebral cortex and hippocampal CA 1 section.Conclusion:The pretreatment of cerebral ischemia has a protective effect on the neurons of the cerebral cortex and hippocampal CA 1 section and can inhibit their neurons from apoptosis.
