摘  要:目的 研究人卵巢癌细胞PCDGF的表达 ,探讨其与卵巢癌恶性表型的关系。方法 应用半定量RT PCR和Westernblot分别检测 3株卵巢癌细胞中PCDGFmRNA和蛋白表达水平。单层细胞增殖实验测定 3株卵巢癌细胞的增殖能力 ;Boy den小室体外侵袭实验测定不同细胞株的侵袭能力。结果  3株卵巢癌细胞PCDGF转录和翻译水平呈同步变化 (P <0 .0 5 ) ,均为SW 6 2 6最高 ,A2 780稍低 ,Skov 3最低。而其增殖、侵袭能力亦为SW 6 2 6最强 ,A2 780次之 ,Skov 3最弱。结论  3株卵巢癌细胞PCDGFmRNA和蛋白表达水平与其增殖、侵袭能力呈正相关 (前者r1=0 .97,r2 =0 .89,后者r1=0 .85 ,r2 =0 .84 ) ,提示PCDGF在卵巢癌发生演进中可能作为独立因素起多种作用 ,有望成为治疗的新靶点。Objective To investigate the expression of PCDGF and to discuss the correlation between the levels of its expression and malignant phenotype in human ovarian cancer cells. Methods Expressions of PCDGF in three ovarian cancer cells were detected by quantitative RT-PCR and western-blot assay. Cell growth assay and Boyden chamber in vitro invasion assay were used to evaluate the capability of both proliferation and invasion in those cells, respectively. Results Levels of PCDGF transcription and translation were coincident in three ovarian cancer cells ( P <0.05). Moreover, the capability of both proliferation and invasion also show the trend: SW626 > A2780 > Skov-3. Conclusion There is a strong correlation between the levels of PCDGF mRNA expression and the capability of both proliferation and that of invasion in ovarian cancer cell ( r 1 =0.97, r 2 =0.89), so did the protein of PCDGF ( r 1 =0.85, r 2 =0.84).PCDGF may play multiple roles in the process of ovarian cancer occurrence and development as an independent factor, and may become new molecular target for epithelial cancer.
