摘  要:目的 :探讨乳腺肿瘤端粒酶活性及hTERTmRNA检测的临床意义。方法 :采用ELISA -TRAP法检测了 4 8例乳腺癌、36例乳腺增生及 30例乳腺纤维腺瘤组织端粒酶活性 ;同时采用RT -PCR检测其hTERTmRNA表达。所有样本行常规病理学检查。结果 :乳腺癌、乳腺增生及乳腺纤维腺瘤组织端粒酶活性阳性率分别为 89.6 % (4 3/ 4 8)、33.3% (12 / 36 )及 2 6 .7% (8/ 30 ) ,hTERTmRNA表达阳性率分别为 91.7% (4 4 / 4 8)、36 .1% (13/ 36 )及 2 6 .7% (8/ 30 )。结论 :端粒酶的激活与乳腺癌的发生密切相关 ,端粒酶活性及hTERTmRNA表达水平的检测有助于乳腺癌的诊断及鉴别诊断。Objective: To study the clinical significance of telomerase activity and human telomerase reverse transcriptase (hTERT) mRNA expression detection for breast tumors. Methods: Telomerase activity was detected by ELISA-based telomeric repeat amplification protocol (ELISA-TRAP) assay in 48 breast cancer,36 mammary hyperplasia and 30 breast fibroadenoma tissues. Expression of hTERT mRNA was also measured by reverse transcriptase-polymerase chain reaction (RT-PCR) assay in these samples. Results: The positive rates of telomerase activity in breast cancer,mammary hyperplasia and breast fibroadenoma tissues were 89.6%(43/48),33.3%(12/36)and 26.7%(8/30),respectively,and those of hTERT mRNA expression were 91.7%(44/48),36.1%(13/36) and 26.7%(8/30),respectively. Conclusion: The telomerase reactivation is significantly correlated with mammary tumorigenesis. Detection of telomerase activity or hTERT mRNA expression may be useful for differential diagnosis of malignant and benign breast lesions.
