摘 要:目的 研究反式二氢二醇环氧苯并芘 [Anti 7,8 dihydrodiol 9,10 epoxidebenzo(a)pyrene ,BPDE]诱导的恶性转化的人支气管上皮细胞 (Humanbronchialepithelialcells ,HBE)及其裸鼠成瘤细胞中DNA断裂损伤情况。方法 单细胞凝胶电泳法 (Singlecellgelelectropheresis ,SCGE)。结果 反式BPDE诱导的恶性转化的HBE及其裸鼠成瘤细胞与对照组细胞相比各指标差异均有显著性 (P <0 0 1) ,各指标的高剂量与低剂量组之间差异也有显著性 (P <0 0 5 )。结论 反式BPDE可导致DNA断裂损伤 ,其损伤程度与剂量有一定关系。Objective To study DNA breakage in the transformed human bronchial epithelial(HBE) cells induced by anti-BPDE[the metabolite of benzo(a)pyrene] and the tumor cells of naked rat developing from the transformed HBE cells. Methods Single Cell Gel Electropheresis(SCGE) assay. Results There were significant differences in all six parameters in the transformed HBE cells induced by anti-BPDE and the tumor cells of naked rat developing from the transformed HBE cells compared with the controlled(P<0.01). There were also significant differences in the total six parameters between the low-dose and the high-dose administrated cells( P<0.05). Conclusions Anti-BPDE can result in DNA strand breakage, and the degrees of damage is related to the administrated doses of anti BPDE.
