摘 要:目的:研究重组内皮抑素对碱烧伤诱导角膜新生血管(CNV)的抑制作用。方法:采用碱烧伤法诱导兔CNV形成。在碱烧伤后的第1天,分别在球结膜下注射50μg/ml重组内皮抑素和生理盐水各0.2ml,隔日1次,共8次,动态观察CNV的生长状况。利用免疫组化的方法检测血管内皮生长因子(VEGF)的表达,并在显微镜下进行微血管计数。结果:重组内皮抑素治疗组CNV的长度和面积、VEGF的表达及微血管数量均明显少于生理盐水对照组(P<0.05)。结论:重组内皮抑素可有效抑制CNV的生长,为临床上碱烧伤引起CNV的预防和治疗提供了新的方法。Objective: To study the inhibitory effect of recombinant endostatin on corneal neovascularization. Methods: Alkali burn was applied to induce the formation of corneal neovascularization(CNV) in rabbits. On the first day after alkali burn, 50靏/ml recombinant endostatin (0.2ml) and normal saline (0.2ml) were respectively injected into subconjunctival tissues of rabbits,one time every other day, eight times in total. The growth state of CNV was observed, the expression of VEGF in cornea was detected with immunohistochemical assay, and the microvessel quantity was counted as well. Results: Endostatin treatment group showed shorter length of blood vessels and smaller area of blood vessels than normal saline group(control group) (P<0.05).The expression of VEGF and microvessel quantity in endostatin treatment group were also significantly lower than those in control group (P<0.05).Conclusion: Recombinant endostatin could effectively inhibit the growth of corneal neovascularization induced by alkali burn, which will provide a new method for the prevention and treatment of corneal neovascularization in clinic.
