Objective: To study the relations among Leptin, Orexin and related hormones of differential obese rats induced by high-fat diet. Methods: Sixty male rats whose weight was between 85g and 95g were weaned for a week and divided into four groups, control group, mild, moderate and severe obese groups. There were 15 rats in each group and their blood levels of Leptin, OrexinA, insulin, growth hormone and TC, TG, HDL-C were measured. Results: The Leptin level decreased but the OrexinA and insulin level of serum increased with the increase of weight. The GH of serum didn't show regulative change with the change of weight. The Orexin, insulin and TC, TG, LDL-C of serum positively correlated with body weight and Lee index, but they negatively correlated with Leptin level. The Orexin and insulin levels of serum positively correlated with the levels of TC, TG, LDL-C. The levels of Leptin, OrexinA, insulin and TC, TG, LDL-C had no association with the level of growth hormone(GH) level. Conclusion: High-fat diet may contribute to obesity by reducing Leptin and improving Orexin and insulin, so recombinant Leptin may have a potential prospect in treatment of obsity.
