摘  要:目的 观察脑出血急性期血肿周边脑组织神经肽Y(neuropeptideY ,NPY)含量的动态变化 ,探讨NPY在脑出血后继发性脑损害中的作用。方法 Wistar大鼠 70只 ,采用抽签法分为手术组及假手术对照组 ,每组各 3 5只 ,每组按出血前 ,出血后 3 0min、6、12、2 4、48、72h分为 7组 ,每小组 5只。采用胶原酶和肝素联合注入尾状核的方法建立大鼠脑出血模型 ,分别于上述各个时相点测定血肿周边NPY的含量、脑含水量。结果 脑出血后 6h血肿周边脑组织NPY的含量开始升高 ,并于2 4h达峰值 ,48h开始回落 ,但至 72hNPY的含量仍显著高于出血前 (P <0 .0 1)。与此同时 ,血肿周边脑组织含水量也同步升高。结论 脑出血过程中 。Objective To observe the dynamic changes of neuropeptide Y (NPY) in perihematoma during acute experimental intracerebral hemorrhage (ICH) and to investigate the roles of NPY in secondary brain damage. Methods A total of 70 Wistar rats were randomized into two groups: control group ( n =35) and ICH group ( n =35). Each group was subdivided into 7 groups at preoperative and postoperative 0.5, 6, 12, 24, 48, and 72 h ( n =5 in each group), respectively. The ICH model was established by injecting collagenase and heparin into the rat caudate nucleus. The changes of NPY and cerebral water contents in perihematoma were observed at preoperative and postoperative 0.5, 6, 12, 24, 48, and 72 h, respectively. NPY contents were determined by radio immunoassay and cerebral water contents by dry/wet weighing method. Results NPY activity and cerebral water content in perihematoma increased synchronously at 6 h, and peaked at 24 h, and then began to reduce at 48 h, and were still higher at 72 h after ICH than those before hemorrhage ( P <0.01). Conclusion NPY may play an important role in the pathogenesis of secondary brain damage during cerebral hemorrhage.
