补肾合剂对牙周炎大鼠外周血中IL-1β含量的影响及牙周组织形态学的改变

摘　　要：目的观察中药补肾合剂对牙周炎大鼠外周血中白细胞介素 1 β(interleukin 1 β ,IL 1 β)水平的影响 ,探讨其治疗牙周炎的机理。方法选用 6 0只 1 0月龄SD大鼠 ,随机分成 6组 ,每组 1 0只。A组为正常对照组 ,B～F组建立大鼠牙周炎模型 ,其中B组为造模对照组 ,C、E组为补肾合剂治疗组 ,C组治疗 4周处死 ,E组治疗 1 2周处死 ,D、F组为补肾合剂治疗对照组 ,分别与C、F组同时处死。采用酶联免疫法测量血清中IL 1 β水平 ,同时观察各阶段大鼠的生活状况及牙周炎造模处牙周组织形态学改变。结果补肾合剂治疗组大鼠外周血中IL 1 β含量在实验的各阶段均明显低于对照组 ,而且治疗组大鼠的全身状况及牙周组织均好于相应对照组。结论补肾合剂可以降低实验性牙周炎大鼠外周血中IL 1 β含量 ,从而进一步改善牙周局部及全身状况。ObjectiveTo observe the effect of kidney benefit Chinese medicine on the level of cytokine interleukin 1β(IL 1β) in the peripheral blood during experimental periodontitis for its treating mechanism. MethodsSixty 10 month old SD rats were divided into six groups at random. Group A was killed at the beginning of the experiment and used as the normal control. Group B-F were used as experimental periodontitis models. The rats in group B were killed at the end of experimental periodontitis establishment to assess the features of the model. Group C and Group E received the therapy of the Chinese medicine for 4 and 12 weeks, and were killed respectively. Group D and Group F, as the control groups, were fed running water as placebo and killed at same time with group C and E respectively. The levels of IL 1β in the peripheral blood were measured with enzyme linked immunosorbent assay after the rats were killed. ResultsThe levels of IL 1β in treated groups were much lower (P< 0.01 ) than those in the control groups (Group D and Group F). At the same time, the general conditions and pathological changes of the experimental groups were better in treated groups than those in the control groups. ConclusionThe kidney benefit Chinese medicine lowered the levels of IL 1β in the peripheral blood of the experimental periodontitis models.
