摘 要:采用钼酸铵定磷法测定不同剂量的乙醇、乙醛作用下红细胞膜Na+ K+ ATPase、Mg2 + ATPase、Ca2 + ATPase活性的变化 ,探讨乙醇、乙醛的细胞膜毒性及其机制。结果显示随着乙醇、乙醛剂量的增加 ,细胞膜Na+ K+ ATPase、Mg2 + ATPase、Ca2 + ATPase酶活性逐渐降低 ,呈现剂量 效应关系。当乙醇剂量为 2mmol/L时 ,各实验组与对照组比差异有显著性 ,而乙醛剂量为 1mmol/L时 ,各实验组与对照相比差异已呈现显著性。表明乙醇、乙醛对细胞膜Na+ K+ ATPase、Mg2 + ATPase、Ca2 + ATPase酶活性均有抑制作用。乙醛是乙醇在机体内的重要代谢产物 ,在研究乙醇的毒性作用及机制时乙醛的作用不容忽视。The effects of alcohol and acetaldehyde on the activities of Na + -K + -ATPase,Mg 2+ -ATPase and Ca 2+ -ATPase in hu-man erythrocyte membrane were investigated in vitro.The activities of these ATPase in the erythrocyte membrane treated with different concentrations of alcohol and acetaldehyde(0,1,2,3and4mmol/L)were determined using spectrophotometry.The results showed that the activities of those three kinds of ATPase decreased with the increase of alcohol and acetaldehyde doses in a reverse dose-effect pattern.At a dose of alcohol≥2mmol/L and of acetaldehyde≥1mmol/L,the activities of Na + -K + -ATPase,Mg 2+ -ATPase and Ca 2+ -ATPase were inhibited obviously,significantly different from those in the control group,indicating that alcohol and acetaldehyde could inhibit the activities of Na + -K + -ATPase,Mg 2+ -ATPase and Ca 2+ -ATPase in the erythrocyte membrane.Acetaldehyde is an im-portant metabolite of alcohol in the bodies,therefore,it should not be ignored that the role of acetaldehyde in studying toxicity and mechanism of alcohol.
