摘  要:目的:观察迷走神经刺激(vagusnervestimulation,VNS)对红藻酸致痫大鼠海马CA1区Ⅰ组代谢型谷氨酸受体(metabotropicglutamatereceptor,mGluR)的影响,为VNS抑痫机制研究提供理论依据。方法:SD大鼠64只数字表法随机分为对照组、红藻酸组、单纯VNS组和VNS治疗组4个组,每组16只,每组再分为测MGluR58只和测mGluR128只。观察大鼠的行为改变,并在各组动物存活2h后,断头取脑,在前囟至前囟后5mm区域内取脑组织用ABC法行mGluR免疫组织化学染色。光镜下以显微计数尺计数双侧海马CA1区1mm范围内阳性神经元的变化。结果:红藻酸组出现重型发作表现;VNS治疗组仅呈轻型发作表现;与对照组相比,红藻酸组CA1区mGluR1a为(50±8)/mm2,mGluR5为(125±9)/mm2,较对照组(42±3),(98±6)/mm2表达增加,差异有显著性意义(q=-4.37,-10.88,P<0.05,0.01),VNS治疗组中mGluR1a表达无明显变化,mGluR5表达显著降低,为(115±8)/mm2。结论:VNS可有效控制癫痫发作,并降低痫后mGluR,特别是mGluR5的表达,提示抑制mGluR的表达可能是VNS发挥抗癫痫作用的重要机制之一。AIM:To observe the effect of vagus nerve stimulation(VNS)on the group I metabotropic glutamate receptor mGluR)after vagus nerve stimulation(VNS)in rat with refractory epilepsy induced by kainic acid(KA),so as to provide theoretical evidence for the mechanism of VNS in inhibiting epilepsy. METHODS:A total of 64 male SD rats were randomized into two groups,and each group was subdivided into control group,KA group,VNS group and KA+VNS groups,so totally there were 8 subgroups of 8 rats in each.The behavioral changes of rats were observed,then their heads were cut down for brain 2 hours after survival,brain tissues were taken from the region from the frontal fontanelle to 5 mm behind it,and the ABC method was performed for immunohistochemical staining.The changes of positive neurons in the range of 1 mm of bilateral hippocampi CA1 area were counted by using micro-ruler under light microscope. RESULTS:There were severe manifestations in the KA group,but only mild manifestations in the VNS group.The expressions of mGluR1a[(50±8)/mm2]and mGluR5[(125±9)/mm2]in the CA1 area in the KA group were significantly increased as compared with those in the control group[(42±3),(98±6)/mm2](q=4.37,-10.88,P< 0.05, 0.01).The expression of mGluR1a did not change obviously, while that of mGluR5 [(115±8)/mm2] was decreased significantly in the VNS group. CONCLUSION:VNS can effectively control the epileptic seizure, and decrease the expression of group I mGluR after epilepsia,especially mGluR5,which indicates that group I mGluR plays important roles in the anticonvusant effect of VNS.
