摘  要:目的:探讨内给氧改善大鼠脑组织缺血再灌注后能量代谢障碍,起到脑保护作用的机制。方法:30只Wistar大鼠随机分成3组:假手术组、缺血组、治疗组,制备大鼠脑缺血再灌注损伤模型,予内给氧治疗后,分别测定各组大鼠脑组织中的三磷酸腺苷(ATP)、二磷酸腺苷(ADP)、单磷酸腺苷(AMP),乳酸含量及Na+-K+-ATPase活性,计算能量负荷值。结果:内给氧治疗组的ATP含量(2.3309±0.0866)mg/L、能量负荷值为0.0960±0.0052,Na+-K+-ATPase活性为(0.903±0.117)μkat/g,均高于缺血组犤(0.8199±0.0591)mg/L,0.1663±0.0044,(0.465±0.064)μkat/g犦差异存显著性意义(P<0.05),乳酸含量(16.0342±1.0136)mmol/g低于缺血组(24.2513±4.3571)mmol/g,差异有显著性意义(P<0.05)。结论:内给氧可明显改善大鼠脑缺血再灌注后能量代谢,具有脑保护作用。AIM:To explore the effect of intra-oxygenation on the improvement of energy metabolic disorder after ischemia reperfusion in rats,and to study its mechanism for protecting brain. METHODS:Thirty Wistar rats were randomly divided into 3 groups: sham operated group,ischemia group and treatment group.Rat models of ischemia reperfusion were established,and received the therapy of intra-oxygenation,then the levels of adenosine triphosphate(ATP),adenosine diphosphate(ADP),adenosine monophosphate(AMP),lactic acid(LA)and the activity of Na+-K+-ATPase were determined,and energy charge of brain was calculated in all groups.RESULTS:The ATP level[( 2.330 9±0.086 6) mg/L], energy charge(0.096 0±0.005 2)and Na+-K+-ATPase activity[(0.903±0.117) ukat/g] in the treatment group were all higher than those in the ischemia group[(0.819 9 ±0.059 1) mg/L, 0.166 3±0.004 4 and(0.465±0.064) ukat/g], and there were significant differences (P< 0.05); and the LA level in the treatment group [(16.034 2±1.013 6) mmol/g] was significantly lower than that in the ischemia group [(24.251 3±4.357 1) mmol/g] (P< 0.05). CONCLUSION:Intra-oxygenation therapy can improve the energy metabolism of brain after ischemia reperfusion of rats,and has the protective effect on brain.
