摘  要:目的:通过雄激素对乳腺癌细胞钠碘转运体(sodium-iodidesymporter,NIS)基因表达的影响,探讨乳腺肿瘤组织NIS的表达特点和调控因素。方法:乳腺癌细胞MCF-7进行培养,分别给予不同浓度的雄激素包括睾酮、双氢睾酮和脱氢表雄酮等刺激72h。分别采用反转录多聚酶链式反应(reversetranscriptase-polymerasechainreaction,RT-PCR)方法和Western方法检测细胞中NIS的mRNA和蛋白表达情况。采用125I测定细胞的摄碘功能。结果:乳腺癌细胞MCF-7在经过睾酮刺激后,细胞中NIS的mRNA和蛋白表达与对照组比较差异有显著性意义(t=1.812,P<0.05),并且随睾酮浓度增高而增加。经过睾酮刺激的细胞摄碘能力增加。而经过双氢睾酮或脱氢表雄酮刺激的乳腺癌细胞中NIS的mRNA和蛋白表达以及摄碘能力没有明显变化。结论:睾酮对乳腺癌细胞MCF-7的NIS基因表达具有调节作用,并影响其摄碘能力。AIM:To investigate the character of expression and adjusted factor of tumor tissue of mammary cancer sodium iodide symporter(NIS)through androgen on gene and protein expression of NIS. METHODS:The breast cancer cells MCF7 were cultured and stimulated with the androgen at different densities including andrusol,anaboleen and dehydroepiandrosterone for 72 hours.mRNA and the albumen expression of NIS in cells were detected by RT PCR method and Western method.And the activity of iodide was assessed by125I . RESULTS:mRNA and albumen expression of NIS in cells were obvious increased and the difference was significant compared with control group after the mammary cancer cells MCF7 were stimulated by andrusol (P< 0.05, t=1.812),which were increased with the increase of the density of andrusol.NIS could been increased in the cells that were stimulated by andrusol.MRNA,the albumen expression and NIS in mammary cancer cells did not obvious change after stimulated by anaboleen and dehydroepiandrosterone. CONCLUSION:Testosterone may play a role in iodide uptake modulating in breast cancer cells by the effect on NIS gene and protein expression.
