大肠癌患者肿瘤细胞及外周血淋巴细胞中P-糖蛋白表达的研究

摘　　要：目的　探讨 37例大肠癌细胞及外周血淋巴细胞 (PBL)P 糖蛋白 (P gp)检出率、表达P gp癌细胞及淋巴细胞的百分率和两者的相关性 ,以及影响P gp表达的因素。方法　免疫组化S P法及流式细胞计数 (FCM )检测 p gp。结果　 37例大肠癌FCM检测结果 :大肠癌细胞P gp总检出率 97.3% ,P gp平均表达率 (7.6 6 3± 1.886 ) %。外周血淋巴细胞P gp总检出率81.1% ,平均表达率 (1.912± 0 .2 4 7) %。免疫组化S P法肿瘤组织P gp总检出率 6 4 .8%。S P法与FCM的阳性符合率 6 6 .6 %。 结论　(1)大肠癌PBL的P gp检出率及肿瘤内的P gp表达率与肿瘤的病理分型、分化程度、Dukes分期有关 ,而与肿瘤的所在部位及性别无关。 (2 )FCM检测P gp比免疫组化S P法准确 ,它能定性定量反映肿瘤内P gp表达情况。 (3)肿瘤组织与外周血淋巴P g表达趋势基本一致 ,具有较好的相关性。Object To investigate the positive expression of P-glycoprotein(P-gp) and the percentage of P-gp positive cells within tumor and peripheral blood lymphocytes (PBL) of the patients with colorecta l carcinoma,and factors which affect their P-gp expression.Methods Using immunohistochemical streptavidin-peroxidase (s-p) technique and flow cytometry assay(FCM) to detecte P-gp.Results The P-gp expression of 37 cases with colorectal carcinoma were 97.3% detected by FCM in tumor tissues(with the average value 7.663±1.886%),81.1% in PBL(with the average value 1.912±0.247%),and 64.8% by S-P method.There were 66.6% corresponded between FCM and S-P.Conclusion (1) The P-gp expression of colorectal carcinoma correlates with its histologic type,differential degree,Dukes′ stage and the age of patient,but has not relation to the position of carcinoma and the sex of patients.(2) FCM is more accurate than S-P,because it quantitatively reflect percentage of P-gp positive cells of tumor tissue.(3) P-gp expression between tumor tissue and PBL behaves the same tendency.
