摘  要:目的 :通过免疫组化方法对视紫质 (rhodopsin)、胶质纤维酸性蛋白 (glialfibrillaryacidicprotein ,GFAP)、波形蛋白(vimentin)、血管内皮细胞生长因子 (vascularendothelialgrowthfactor,VEGF)的检测 ,从分子水平观察光动力疗法 (PDT)治疗后非脉络膜新生血管形成 (CNV)区的视网膜的蛋白表达情况 ,对PDT的安全性进行研究。方法 :采用过量的眼底激光破坏Bruch膜 ,建立猴眼CNV模型 ;将正常猴眼作为对照 1组 ,未作PDT治疗的CNV模型眼作为对照 2组 ;通过免疫组化的方法来研究PDT治疗后非CNV区 1、2、3、6个月的猴眼视网膜的视紫质抗体、胶质纤维酸性蛋白抗体、波形蛋白抗体、血管内皮细胞生长因子抗体及其原位杂交 ,检测PDT治疗组与对照组的视网膜抗体表达情况。结果 :猴眼视网膜非CNV区各层组织经PDT治疗后与对照组相比视紫质抗体、胶质纤维酸性蛋白抗体、波形蛋白抗体、血管内皮细胞生长因子抗体及血管内皮细胞生长因子原位杂交信号均未见明显差异。结论 :PDT治疗后并未引起视网膜各层蛋白表达异常 ,VEGFmRNA在分子水平亦未见表达改变 ,因此PDT治疗对视网膜组织是安全的。Objective:To evaluate the safety of photodynic therapy(PDT) in the retina without choroidal neovascularisation(CNV) following PDT at molecular level by observing the expression of several related factors (rhodopsin, glial fibrillary acidic protein (GFAP), vimentin, vascular endothelial growth factor (VEGF)) with immunohistochemistry. Methods:CNV monkey model was successfully established by destroying Bruch's membrane with intense laser,and normal monkeys were secleted as control groups. The expressions of rhodopsin, GFAP, vimentin and VEGF in the non CNV retina after PDT were studied with immunohistochemistry and in situ hybridization at 1,2,3 and 6 month and compared with control groups. Results:No significant differences of rhodopsin, GFPA,vimentin and VEGF expression were detected between the non CNV retina following PDT and the control groups.Conclusion:No abnormal protein expressions are observed histopathologically in retinas following PDT, which suggests that PDT might be safe modality for retinal tissue.
