摘 要:本文对水煮法和醇提法两种不同提取苜蓿多糖方法以及蒽酮-硫酸法和苯酚-硫酸法两种不同的苜蓿多糖测定方法进行了比较。结果发现,在进行苜蓿粗多糖的提取时,利用水煮法提取粗多糖可显著提高多糖得率。用蒽酮-硫酸法和苯酚-硫酸法两种不同的测定方法来测定多糖含量,在未脱蛋白质前,两种测定方法皆具有良好的稳定性、精密性以及重现性,但都受到样品中蛋白质干扰严重;而在脱蛋白后,蒽酮-硫酸法在检测样品不同稀释度的重现性稍优于苯酚-硫酸法。Two extraction methods and two assay methods were applied to analy si s polysaccharides in alfalfa. As a result, between the two extraction methods of boiling water-extraction and alcohol-precipitation to separate polysaccharide s from alfalfa, water-extraction method is more suitable than that of alcohol-precipitation. Between the two spectrophotometric methods of anthrone-sulfuric acid and phenol-sulfuric acid for polysaccharide determination, it was discover ed that both methods present same reproducibility before removing protein in the sample. But after removal of protein in the sample, the reproducibility of anth rone-sulfuric acid spectrophotometric method is better than that of phenol-sul furic acid spectrophotometric method.
