摘  要:目的 动态研究Bax和bcl-2在树鼩肝癌形成过程的不同阶段的表达与肝细胞凋亡的关系。方法 用免疫组化及TUNEL方法,检测35例树鼩的第30、60周肝活检组织和肝癌组织及对照树鼩对应期肝活检组织中Bax、bcl-2表达和细胞凋亡指数(AI)。结果①Bax和bcl-2在肝癌组织中的表达率均显著高于癌前和空白对照同期肝组织;在60周的肝组织中也显著高于30周及空白对照组对应期肝活检组织。②癌组织及其第30和60周的肝组织中的AI均值均显著高于对照组对应期肝组织。肝癌组织与其癌前30及60周的肝活检组织之间的AI差别无显著性。③Bax和bcl-2阳性肝组织的凋亡指数也均显著高于阴性的。结论 1.正常树鼩肝细胞凋亡率极低,在肝癌形成过程中凋亡率增加;2.Bax和bcl-2都与树鼩肝癌的发生有关,在肝癌形成过程中,它们的表达率及强度均呈增加趋势。动态观察树鼩肝癌形成过程中一些基因的表达变化,有利于探讨肝癌的发病机理,为人类肝癌的早期诊断、治疗以及预防的研究提供线索。肝组织中的Bax、bcl-2基因表达状况对于预测肝癌的发生可能有一定帮助。Objective To research the relationship between the expression of Bax and bcl-2 and liver cell apoptosis in different stages during hepatocarcinogenesis of tree shrews. Methods The expression of Bax and bcl-2 in the tissues of liver and hepatocellu lar carcinoma (HCC) were detected with immunohistochemistry. TUNEL technique was used to determine the apoptosis cells in all the samples mentioned above. Results (1) The expression rate of Bax and bcl-2 in liver cancer were significantly higher than that in biopsy samples at 30 th, 60 th week and the control animals, they were also significantly higher in biopsy ones at 60 th week than that in 30 th week and the control ones. (2) The apoptosis indexes in HCC and tissue biopsies at 30 th and 60 th week were obviously higher than that of the control animals. (3) The apoptosis index in Bax and bcl-2 positive tissues were much higher than that in their negative tissues. Conclusion (1) The rate of apoptosis is very lower in normal tree shrews liver cells and it increased during hepatocarcinogenesis. (2) The expressions of Bax and bcl-2 are gradually increased during tree shrews hepatocarcinogenesis. The present study might be useful to research the mechanim of HCC development. The expression states of Bax and bcl-2 might be helpful for prognosticating HCC.
