摘  要:目的 探讨原发性肝细胞癌 (HCC)中FHIT基因的表达。方法 采用免疫组织化学技术 (SP法 )检测 4 0例HCC及其癌旁组织、32例肝外伤后正常肝组织中FHIT的表达情况。结果  4 0例HCC中有 1 1例FHIT表达缺失 ,而相应的癌旁组织中只有 2例FHIT蛋白表达缺失 ;正常肝组织中全部呈阳性表达 ;HCC组织中FHIT蛋白缺失率显著高于相应癌旁及正常肝组织 (P <0 .0 1 ) ,并与肿瘤的病理学分级、TNM分期、门静脉癌栓及HBsAg阳性表达有密切关系 (P <0 .0 5 )。结论 FHIT蛋白的缺失与HCC的发生。Objective To study whether the FHIT gene is involved in the genesis of primary hepatocelluar carcinoma(HCC).Methods The tumor samples in 40 cases of HCC,the corresponding paraconcerous tissues and the normal liver tissues in 32 cases of liver trauma were examined for the FHIT expression by SP immunohistochemistry of anti FHIT monoclonal antibody. The FHIT protein expression was negative in 11 of 40 tumor samples,whereas in 2 of 40 corresponding paraconcerous tissues,none of 32 normal liver tissues.The FHIT protein deletion in HCC was higher than that in paraconcerous tissues and normal liver tissues( P < 0.01 ).The FHIT protein deletion rate was also correlated remarkably with Edmondson grade of HCC( P < 0.05 ),and associated with TNM staging,the cancerous thrombi within the portal vein and the HBsAg( P < 0.05 ).Conclusion The FHIT protein deletion may play an important role in the carcinogenesis of HCC.
