摘  要:目的 探讨非小细胞肺癌 (NSCLC)患者外周血微转移的分子诊断的临床意义。方法 应用逆转录聚合酶链反应 (RT -PCR)技术 ,对 31例肺癌患者和 10例肺良性病变患者外周血中MUC1基因mRNA表达进行检测。结果 术前 10例NSCLC患者检测到外周血肺癌微转移 ,外周血微转移与肺癌组织学类型 ,细胞分化程度及P -TNM分期均存在密切关系 (P <0 .0 5 )。 10例肺良性病变患者的外周血中未检测到MUC1mRNA表达。结论 MUC1基因mRNART -PCR法检测肺癌患者外周血微转移具有较高的特异性和敏感性 ,可为制定治疗方案和评估预后提供重要参考依据。Objective To investigate the clinical significance of molec ul ar diagnosis of micrometastasis in peripheral blood of patients with non-small -cell lung cancer.Methods The MUC1 mRNA expression was detecte d in peripheral blood from 31 patients with lung cancer and 10 patients with ben ign pulmonary lesion by nested RT-P CR.Results In the 31 lung cancer patients, MUC1 mRNA expression was detected in 10 peripheral blood samples. The micromatastasis in peripheral blood was closely related to the pathological classification, cell differentiati on of the cancer and P-TNM stage of the disease (P<0.05). No expression of MUC1 mRNA was found in the samples from patients with benign pulmonary lesion. Conclusion RT-PCR amplification of MUC1 mRNA is a specific and sensitive method of detecting micromatastasis in peripheral blood of patients w ith lung cancer . It can also provide important clinical information for pro gnosis and selection of appropriate treatment strategies.
