摘 要:目的 研究维拉帕米对牛眼小梁细胞 (BTMC)增殖和吞噬功能的影响。方法 采用MTT和3 H TDR实验检测 0 0 0 0 1、0 0 0 0 5、0 0 0 1、0 0 0 5、0 0 1、0 0 5、0 1mg/ml 7种质量浓度的维拉帕米对BTMC增殖功能的影响。以乳胶微粒为标记 ,定量检测等于和低于 0 0 1mg/ml维拉帕米对BTMC吞噬功能的影响。结果 低于 0 0 1mg/ml维拉帕米对BTMC增殖功能无抑制作用 (P >0 0 5 ) ,等于或高于 0 0 1mg/ml的维拉帕米可浓度依赖性地抑制BTMC增殖功能 (P <0 0 5 ) ,其IC50 分别为 0 0 3 2 3mg/ml(MTT)和 0 0 2 5 9mg/ml( 3 H TDR)。 0 0 0 1、0 0 0 5、0 0 1mg/ml维拉帕米可浓度依赖性地促进BTMC的吞噬功能 (P <0 0 5 )。结论 临床应用的 0 12 5 %维拉帕米滴眼剂在房水中的质量浓度为 ( 160 7± 2 72 )ng/ml ,该质量浓度范围不抑制BTMC增殖。选用等于和低于 0 0 1mg/ml作为后续实验的质量浓度。维拉帕米还可通过促进小梁细胞的吞噬功能 ,减少房水外流阻力 。ObjectiveTo study the effects of Verapamil on the proliferation and phagocytosis of bovine trabecular meshwork cells(BTMC).MethodsThe effects of verapamil on the proliferation of BTMC were assessed by MTT and 3 H TDR at 0 000 1 mg/ml,0 000 5 mg/ml,0 001 mg/ml,0 005 mg/ml,0 01 mg/ml,0 05 mg/ml and 0 1 mg/ml.The effects of Verapamil equal to or less than 0 01 mg/ml on phagocytosis of BTMC was quantitatively measured by using latex beads as markers.ResultsVerapamil less than 0 01 mg/ml had no significant effects on the proliferation of BTMC( P >0 05).Equal to or higher than 0 01 mg/ml of Verapamil inhibited the proliferation of BTMC in a dose dependent manner ( P <0 05).The IC 50 was 0 032 3 mg/ml(MTT)and 0 025 9 mg/ml( 3 H TDR)respectively.0 001 mg/ml,0 005 mg/ml,0 01 mg/ml of Verapamil promoted the phagocytosis of BTMC in a dose dependent manner( P <0 05).ConclusionsA concetration of (1 607±272)ng/ml in aqueous humor is obtained by using the eye drop of 0 125%verapamil.However this concentration of verapamil doesn't inhibit the proliferation of BTMC,the concetration equal to or less than 0 01 mg/ml should be selected in following experiments.In addition,verapamil may be expected to treat POAG by blocking pathogenesis through promoting the phagocytosis of BTMC and decreasing resistance to the outflow of aqueous humor subsequently.
